1.0 OBJECTIVE
To lay down the procedure for Preparation, Sterilization & Storage
of media.
2.0
SCOPE
This SOP shall be applicable for preparation,
sterilization & storage of media
in Microbiology Laboratory.
3.0
RESPONSIBILITY
3.1
Microbiologist
and above shall be responsible for the
preparation of this SOP.
3.2
Sr. Microbiologist shall be responsible for the
checking of this SOP.
3.3
Manager-QA shall be responsible
for the approval of this SOP.

5.0
PROCEDURE
5.1
Ensure for the date of expiry of the media and the
instructions for preparation of media
mentioned on the container shall be read also before using the media.
5.2
The desired quantity of media shall be weighed using
butter paper and dry spatula.
5.3 Weighed media shall be transfer into a clean dry
flask (about 2 times larger than the desired volume).
5.4 The measured volume of distilled water shall be
add in steps from the measuring cylinder and ensure for the stir thoroughly to
dissolve.
5.5 The remaining volume of water shall be add and place
the flask on Hotplate and heat to dissolve the contents (avoid excess heating).
5.6
Thermo-sensitive stickers are to be pasted on
the flask after it is cooled to 45 to 500C.
5.7
Ensure the pH of the media before
sterilization and adjust the pH, if required by using 0.1 M HCL and 0.1 M NaOH.
5.8
The media shall be sterilized as per instruction
of the manufacturer.
5.9
The prepared media shall be labeled with details,
i.e. Name, Date of Preparation, and Use before date & Signature.
5.10
Agar media shall not be re-melted more than
once.
5.11
The media shall be stored at 2 to 80C
in the refrigerator for one month.
5.11Media
boxes shall be used at the FIFO system.
5.12Gloves
and mask shall be used before weighing the media
5.13Frequency:
As and when required.
