SOP FOR MANTRA IMAGE SYSTEM

1.0 OBJECTIVE

To design the procedure for operation of Microplate reader from Envision.

2.0 SCOPE

This Standard Operating Procedure (SOP) is applicable for the operation, maintenance and calibration of Microplate reader.

3.0 RESPONSIBILITY

3.1 All personnel in the Molecular Biology Department shall be responsible for implementation and compliance to the SOP.

3.2 Head/Designees of Molecular Biology department shall be responsible to ensure the implementation and compliance to this SOP.

4.0 DISTRIBUTION

4.1 The Quality Assurance department responsible to keep SOP ‘Master Copy’ approved through Quality Manager.

4.2 The copy of ‘Control Copy’ of SOPs is being distributed in Molecular Biology department and placed near related Equipment/ Instrument as Display copy.

5.0 DEFINITION

5.1 Definition

5.1.1 Microplate reader: The Plate Reader may be used to quickly take optical measurements of many liquid samples in the microliter wells of a single plate

6.0 PROCEDURE

6.1 Principle

The microplate reader is a multimodal instrument that allows for a variety of experiments to be performed and measured simultaneously. Microplate readers can make absorbance, fluorescence and luminescence measurements. Multiwell plates are integral to the microplate reader and allow for many experiments to be performed at once. Regardless of the assay type, experiments on the plate reader utilize a standard curve to determine the experimental values.

6.2 Operations

6.2.1 Turn on the instrument

6.2.2 Wait till initialization is completed

6.2.3 Connect to the EnVision software

6.2.4 Select the Method

6.2.5 The Method can also be edited by clicking EDIT icon and desired parameters can be changed.

6.2.5 Click on the PLATE icon to load the plate in the slot.

6.2.6 Click RUN

6.2.7 Take out the plate once reading is complete.

6.3 Maintenance

6.3.1 The plate carrier should be kept clean to avoid dust and dirt entering into the optics at the measuring position.

6.3.2 In case of spillage of a sample containing any signal generation component, clean the area where the spillage occurred.

6.3.3 The plate carrier should be cleaned at least once a week using a soft cloth or tissue paper soaked in a mild detergent solution or alcohol.

7.0 PRECAUTION

7.1 Instrument should be kept on leveled surface.

7.2.1 Avoid spilling liquids near the instrument.
