1.0 OBJECTIVE
1.1 To lay down a standard procedure to be followed for the Operation, Calibration, and Maintenance of Centrifuge in Animal House Facility.
2.0 SCOPE
2.1 This Standard Operating Procedure (SOP) shall be applicable for Operation, Calibration, and Maintenance of Centrifuge in Animal House Facility.
3.0 RESPONSIBILITY
3.1 Head- Animal House Facility
3.2 All Personnel involved in the usage of Centrifuge.

5.0 PROCEDURE
5.1 Operating Procedure
5.1.1
Plug in the chord of the instrument and turn on the
power of the instrument.
5.1.2
Take appropriate size sterile centrifuge tubes and
collect the sample to be centrifuged.
5.1.3
Adjust the volume/weight of content in the centrifuge
tubes to avoid any breakage/spillage of content during centrifugation
process.
5.1.4
Do not keep the level of the liquid in each tube
more than 2/3 volume of the tube.
5.1.5
Ensure that the tubes are closed properly before
keeping it in the slot.
5.1.6
Open the lid of the chamber.
5.1.7
Place the centrifuge tube in exact opposite slots. Each
and every centrifuge tube should have a centrifuge tube of similar volume /
weight on the opposite side.
5.1.8
Close the lid of the chamber.
5.1.9
Switch ON the mains for cooling and Set the required
temperature.
5.1.10
There is a digital temperature controller for the
selection of temperature.
5.1.11
Press the ‘SET’ button and adjust the temperature by
using Up and Down navigation keys.
5.1.12
Greenlight will glow on the ‘COOLING ON’ button.
5.1.13
Once the required temperature is adjusted press ‘ENT’.
5.1.14
Switch on the mains for ‘RPM’. Adjust the ‘RPM’ with
the help of a knob.
5.1.15
Set the time required by pressing the “SET TIME’ button.
5.1.16
After centrifugation time is over, bring the knob to
minimum position
5.1.17
Switch OFF the mains.
5.1.18
Ensure that motor is stopped & centrifuge tubes
are in a rest position.
5.1.19 Open the lid and remove the tubes.
5.2 Calibration
5.2.1
Calibration of the Centrifuge shall be carried out
by the outside agency.
5.2.2 Calibration shall be done once a year or as per requirement and calibration label shall be updated subsequently.
5.3 Cleaning and Maintenance
5.3.1 Wipe out the centrifuge machine from outside and
inside.
5.3.2 Clean the instrument before & after use every
experiment.
5.3.3 Service the instrument by authorized service agency and service report to be maintained.
Note:- Take care of an overflow of liquid in the chamber.
Place the test tube on the opposite side to
prevent unbalancing during centrifugation.
Increase the speed
slowly.
