 EVALUATION OF SPERMATOGENIC EFFECT OF HERBAL FORMULATION IN EXPERIMENTAL ANIMALS.

1.0  TEST SYSTEM DETAILS:

Species                  : Rattus norvegicus (Rat)

Strain                    : Sprague Dawley

Age                       : 4-6 weeks

Body Wight          : 150-180 g

Sex                        : Male

No. of animals      : 10 M in each group

Total animals        : 50 M + 15F 

2.0   ALLOCATION OF GROUPS

*Route of drug administration will be decided at the time of experiment mainly preferred oral dosing

6 male rats will be taken extra for the randomization & 15 female rats needed to evaluate male sexual behaviors.

3.0  METHOD:

·       Healthy animals will be selected, randomized based on body weight and divided into 5 different groups.

·       Animals will be weighed, numbered, and divided into different groups.

·       The mode of administration will be oral in dose dependent manner by feeding needle for 28 days.

·       Copulatory Behaviour will be evaluated on day 29 in black observation box by introducing male animal with female rats for 30 minutes.

·       Number of mounting, intromission and ejaculation behavior were noted and recorded for all groups.

·      Copulatory behavior is a sexual behavior in which a male animal introduces sperm into a female's body. It consists of mounting, intromission, and ejaculation.

4.0  END POINT PARAMETER(S):

·       Effect on weight of sexual organs and histological studies

·       Effect on male sexual behavior

·       Sperm count

·       Fructose content in seminal vesicles

·       Hormone assay (Testosterone)

·       Histopathology of Testes and Epididymis

·       Number of mounting, intromission and ejaculation behavior (Optional)
