STANDARD OPERATING PROCEDURE FOR VALIDATION OF AUTOCLAVE


1.0 OBJECTIVE:

1.1 To validate the steam sterilization method of autoclave by biological indicators and chemical indicators.

2.0 PROCEDURE:





2.1 By Biological Indicator

2.1.1 Take five strips before sterilization and proceeds for biological indicators performance test as per

2.1.2 Take another five strips of biological indicator and put on different places of autoclave.

2.1.3 Run autoclave at 121° for 1.5 minutes.

2.1.4 Remove the strips and proceed the test for spores count as per

2.1.5 Put again new five strips of biological indicator and run the autoclave at 121° for 15 minutes.

2.1.6 After completion of the autoclave remove the strips of biological indicator and proceed the spores count as per

2.2 By Chemical Indicator

2.2.1 Put the strips of chemical indicator in different places of the autoclave.

2.2.2 Run the autoclave at 121° for the 15 minutes.

2.2.3 After completion of autoclave remove all the strips and check the color of the indicator.

2.2.4 Color of the indicator should change pink to green.

2.3 Procedure:

2.3.1 By Biological Indicator

2.3.2 Take five strips before sterilization and proceeds for biological indicators performance test as per SOP/MICRO.....

2.3.3 Take another five strips of biological indicator and put on different places of Oven.

2.3.4 Run oven at 160°C for 3 minutes.

2.3.5 After completion of the cycle remove all the strips and proceed the test for spores count as per SOP/MICRO......

2.3.6 Put again new five strips of biological indicator and run the oven at160°C for 2 hours for

2.3.7 After completion of the cycle remove all the strips of biological indicator and proceed with the spores count as per SOP/MICRO......


2.3.8 By Chemical Indicator

2.3.9 Put the strips of the chemical indicator in different places of the oven.

2.3.10Run the oven at 160°C for 2 hours

2.3.11 After completion of the cycle remove all the strips and check the color of the indicator.

2.3.12 Color of the indicator should change from tan to black.
