SOP FOR pH METER | CLEANING, OPERATION & CALIBRATION OF pH METER


1.0 OBJECTIVE

To lay down the procedure for cleaning, operation & calibration of the pH meter.

2.0 SCOPE

This SOP shall be applicable for cleaning, operation & calibration of the pH meter in the Microbiology
 Lab.



 

3.0 RESPONSIBILITY

3.1 Microbiologists and above shall be responsible for the preparation and execution of this SOP

3.2 QC & QA Head shall be responsible for the checking and approval of this SOP.

5.0 PROCEDURE

5.1 Cleaning

5.1.1 Ensure that the power supply to the pH meter is switched ‘OFF’ before cleaning and remove the plug from the socket.

5.1.2 pH meter shall be kept on a clean & vibration-free table/platform.

5.1.3 A platform of the pH meter shall be wiped with a dry lint-free cloth on a daily basis so that there will be no dust particles on the pH meter.

5.2 Operation

5.2.1 Ensure the cleanliness of the instrument.

5.2.2 Ensure that the electrode is connected to the pH meter.

5.2.3 Ensure that the power supply switch is “ON” on the pH meter.

5.2.4 Ensure that all the solutions are at the specified temperatures.

5.2.5 Temperature shall be set with the help of the thermostat control knob at the desired temperature.

5.2.6 Set the function selector switch at “STAND BY” (Ensure that the function selector switch is at“STAND BY” between different pH measurements).

5.2.7 Ensure that the digital display reads “0.00” at the “STAND BY” position.

5.2.8 Electrode shall be washed with distilled water and dried with tissue paper.

5.3 Calibration Procedure

5.3.1 Electrode shall be a dip in a 7 pH buffer solution.

5.3.2 “TEMPERATURE 0C” control shall be set to the buffer solution temperature.

5.3.3 Ensure the “STAND BY” switch is “ON”.

5.3.4 7 pH shall be adjusted with “CALIBRATE” control till the digital display shows the

precise pH value of the 7pH buffer solution.

5.3.5 Ensure for the switch “STAND BY” is off and the electrode shall be removed from the buffer solution & wash it with distilled water.

5.3.4 The electrode shall be a dip in a 4 pH buffer solution.

5.3.5 The above procedure shall be repeated for 4pH buffer solution as per 7pH buffer solution.

5.4 pH Measurement

5.4.1 The electrode shall be a dip in solution under test.

5.4.2 Ensure for the switch “STAND BY” is ON.

5.4.3 The display shows the pH value of the solution directly in the pH units.

5.4.4 Then switch to “STAND BY” off and the electrode shall be removed from the solution & wash it with distilled water.
