The purpose of the document is to establish a general and uniform method for the determination of optical density.
2.0 SCOPE
This standard testing procedure is suitable for the determination of optical density in the honey sample.
3.0 RESPONSIBILITY
The expert and trained chemist are responsible for following this procedure. The technical manager is responsible for implementing this procedure.
4.0 REQUIREMENTS
4.1 Apparatus
4.1.1 Spectrophotometer ( 660 nm)
4.2 Glassware: NA
4.3 Reagent: NA
5.0 PROCEDURE
5.1 Weigh accurately 2 g of honey sample in a beaker & dissolve it in distilled water
5.2 Make up the solution with water in a 10 ml measuring cylinder.
5.3 Adjust the spectrophotometer with distilled water in a cuvet at ‘0’ absorbance at 660nm.
5.4 Take the honey solution in the cuvet and read the absorbance directly or as the percent transmittance at the same wavelength.
6.0 CALCULATION
Optical Density = Absorbance
