Purpose:
This
Standard Operating Procedure (SOP) outlines the proper and safe method for
disposing of microbial cultures to prevent the release of potentially harmful
microorganisms into the environment.
Scope:
This
SOP applies to all personnel involved in handling and disposing of microbial
cultures within the facility.
Responsibilities:
a. Laboratory personnel:
Follow
this SOP for proper disposal of microbial cultures.
Ensure
compliance with safety protocols and regulations.
b.
Laboratory supervisor:
Provide
necessary training on disposal procedures.
Oversee
the implementation of this SOP.
Materials
and Equipment:
a.
Autoclave or alternative sterilization method (if applicable).
b.
Biological safety cabinets (BSC) or laminar flow hoods.
c.
Chemical disinfectants.
d.
Appropriate personal protective equipment (PPE) such as gloves, lab coats, and
eye protection.
Procedure:
a. Step 1: Identification and classification
Identify
the microbial cultures to be disposed of.
Determine
the appropriate classification of the cultures (e.g., Biosafety Level 1, 2, 3,
or 4) according to institutional guidelines and regulations.
b. Step 2: Inactivation and decontamination
For Biosafety Level 1 culture:
Transfer
the cultures to a biosafety cabinet or laminar flow hood.
Add
an appropriate disinfectant directly to the culture, following the
manufacturer's instructions for concentration and contact time.
Allow
the disinfectant to act for the recommended duration.
Mix
the culture gently to ensure thorough inactivation.
Decontaminate
all used equipment and surfaces with a suitable disinfectant.
Dispose
of the inactivated cultures as directed in Step 3.
For Biosafety Level 2, 3, or 4 cultures:
Transfer
the cultures to a biosafety cabinet or laminar flow hood.
Follow
the appropriate containment procedures and guidelines for the specific
biosafety level.
Inactivate
the cultures using approved methods, such as autoclaving, incineration, or
chemical treatment.
Decontaminate
all used equipment and surfaces with an appropriate disinfectant.
Dispose
of the inactivated cultures as directed in Step 3.
c. Step 3: Disposal
Place
the inactivated microbial cultures in a leak-proof, autoclavable bag or
container labeled for biohazardous waste.
Autoclave
the bag or container according to the facility's sterilization protocols.
Before disposing of the culture media used in microbiological testings first of all collect and wrap the media properly in aluminum foil and then final pack in polyethylene and autoclave the media at 121°C /30 minutes/15psi. After autoclavation drain the liquid media into the sink under running tap water. Soak the empty glassware in the chromic acid solution for 2hr and then wash the glassware and dip them in 70% IPA Solution for 30 minutes and then rinse with tap water and finally rinse with purified water and dry.
Allotment of load/batch number to disposal activity:
Every time when disposal activity is performed a load/batch number must be given. The disposal number must e given in the following manner.
XXX/YYY/ZZZ
Where
XXX indicates the year in which the activity is performed e.g. 2023
YYY indicates the abbreviation of the activity performed. e.g. disposal of used media (DUM).
ZZZ
indicates serial number e.g. (001)
NOTE: Always use
protective gloves, goggles, overalls, and face masks while performing the disposal
activity.
Always
run an autoclavation activity with empty chambers at 121°C /30 minutes/15psi after
the sterilization of the used media.
If
autoclaving is not available, follow institutional guidelines for alternative
disposal methods, such as incineration or off-site disposal by a certified
waste management service.
Dispose
of sterilized waste as directed by local regulations and waste management
procedures.
Documentation and Record Keeping:
Maintain
accurate records of the disposal process, including the date, description of
cultures, disinfection method used, and disposal method.
Keep
records for the recommended duration, as per institutional guidelines and
regulatory requirements.
Training:
All
personnel involved in handling microbial cultures must receive training on this
SOP.
Provide
refresher training periodically and whenever changes are made to the disposal
procedures.
