Standardization of Diabetes Model in Rabbits before Wound Generation
The following points need to be considered before starting a diabetic wound study on rabbits:
1. Rabbits' weight should be more than 2 kg.
2. Provisions for the rabbit run should be arranged. rabbits are kept in rabbit run after diabetes induction.
3. Rabbits' food should not be given in the rabbit run.
4. A single dose of Alloxan monohydrate (150 mg/kg) is needed intravenous infusion route to induce diabetes to New Zealand Rabbits.
5. During diabetes induction with Alloxan there maybe chances of 5 types of variants may present in the diabetes study.
a. Hypoglycemic
b. Hyperglycemic
c. Insulin-induced (I) Hypoglycemic (II) Appetite loss
d. Not Induced
e. Reversed to Non-diabetic
6. Rabbits show a lower Blood Glucose Level (BGL) in the morning and high in the evening & you will find the highest insulin level every third day evening. Initially, since diabetes induction and up to diabetes stabilization in rabbits, it is necessary to take BGL twice a day.
7. 20- 25% animals remain not induced after the first dose of alloxan Injection; need to be re-induced after 48 hours.
8. Animals should be observe up to the first 48 after alloxan induction, there may be chances of hyperglycemic or hypoglycemic shock after the first 6 hrs.
9. Possibility of hypoglycemia is more in rabbits after the first 6 hours of alloxan induction. So it is necessary to take blood glucose level before 15 minutes of 6 hours.
10. In a diabetes study, more than 30 % of animals get die during the induction of diabetes.
11. Rabbits diabetes study takes at least one month to stabilize the blood glucose level of more than 300 mg/dl.
12. 0.2 % Nitrofurazone is the best standard drug for diabetic wounds in rabbits.
13. Some parameters like wound area, % relatively wound area contraction, blood glucose level (BGL), Insulin consumption, epithelialization time, inflammatory phase, proliferation phase, the single dose OGTT, Multiple-dose OGTT by AUC calculation are considered normal during this study.
