1.0 PURPOSE
The purpose of the document is to establish a general and uniform method for the determination of titratable acidity in skimmed milk powder.
2.0 SCOPE
This standard testing procedure is suitable for the determination of titratable acidity in skimmed milk powder.
3.0 RESPONSIBILITY
The expert and trained chemist are responsible for following this procedure. The technical manager is responsible for implementing this procedure.
4.0 REQUIREMENTS
4.1 Apparatus
4.1.1 Analytical balance
4.2 Glassware
4.2.1 Burette-Graduated in 0,l ml and with an accuracy of 0.05 ml
4.2.2 Pipette- 2 ml
4.2.3 Measuring cylinders- 50 ml
4.2.4 Conical flasks-100 ml or 150 ml
4.3 Reagent
4.3.1 Sodium hydroxide: 0.100M solution
4.3.2 Reference color solution: 3.0 %( dissolve 3 g of Cobalt (II) sulfate heptahydrate (CoSO4.7H2O) in water and makeup to 100 ml).
4.3.3 Phenolphthalein solution:
4.3.4 2.0%Dissolve 2 g of phenolphthalein in 75 ml of 95 %( V/V) ethanol and add 20 ml of water. Add the sodium hydroxide solution until 1 drop gives a faint pink coloration and makeup to 100 ml of water.
5.0 PROCEDURE
5.1 Transfer the sample to a clean, dry container (provided with an air-tight lid) of capacity about twice the volume of the sample.
5.2 Close the container immediately, and thoroughly mix the contents by repeatedly shaking.
5.3 Weigh the (500/α+ 0.01g of the test sample into each of two conical flasks, α being the solids-not-fat content of the sample, expressed as a percentage to two decimal places.
5.4 Prepare reconstituted milk by adding 50 ml of water at about 20°C to the test portion and agitating vigorously. Allow standing for about 20 min.
5.5 Add to one of the conical flasks 2 ml of the reference color solution to obtain a color standard, and mix by slight swirling.
5.6 If a determination on similar products (series) is to be carried out, this color standard may be used
throughout. However, it should be discarded after 2 hrs.
5.7 Add 2 ml of the phenolphthalein solution to the second conical flask, and mix by slight swirling.
5.8 Titrate the contents of the second conical flask, while swirling, by adding the sodium hydroxide solution from the burette until a faint pink color similar to that of the color standard persists for about 5 s. The titration should be completed within 45 s.
5.9 Record the volume, in milliliters, of sodium hydroxide solution used, to the nearest 0.05 ml.
6.0 CALCULATION
Titratable
acidity = 2 x V
Where,
V is the volume, in
milliliters, of the sodium hydroxide solution used for titration.
Express the result to one decimal
place.
For calculating the titratable acidity in terms of percentage, the following formula may be used
Percentage titratable acidity (as Lactic acid)
= 0.09x2xV
Where,
V = volume in milliliters of sodium hydroxide solution used for titration.
