SOP FOR THE ANALYSIS OF MERCURY IN THE BONE SAMPLE USING THE ICP-MS ANALYTICAL TECHNIQUE

Standard Operating Procedure (SOP) for the analysis of mercury in the bone of rats and mice using the ICP-MS analytical technique, based on the provided procedure:

1.0 Objective: To determine the mercury content in rat and mouse bone samples using ICP-MS (Inductively Coupled Plasma Mass Spectrometry).

2.0 Materials and Reagents:

Bone sample from Rat and Mouse

HNO3 (Nitric Acid) - 60 ml

H2O2 (Hydrogen Peroxide) - 30 ml

HCl (Hydrochloric Acid) - 20.1 ml

3.0 Procedure:


3.1 Sample Preparation:

Collect bone samples from rats or mice.

Ensure that all samples are free of muscular traces.

3.2 Sample Drying:

Place the bone samples in a drying oven set to 37°C.

Allow the samples to dry overnight to remove any moisture.

3.3 Sample Weighing:

After drying, cut each bone sample into small pieces with an approximate weight of 27-30 mg.

3.4 Sample Digestion:

Prepare a digestion mixture by combining the following acids:

HNO3 (2 ml/sample)

H2O2 (1 ml/sample)

HCl (0.67 ml/sample)

3.5 Digestion Process:

Add 3.67 ml of the digestion mixture to each bone sample.

Place the samples in containers suitable for acid digestion.

Allow the samples to sit overnight while shaking.

3.6 ICP-MS Analysis:

After overnight digestion, prepare the ICP-MS instrument according to the manufacturer's guidelines.

Inject the digested bone samples into the ICP-MS instrument for analysis.

Record the mercury concentration in each sample.

3.7 Safety Precautions:

Perform all steps in a well-ventilated fume hood.

Wear appropriate personal protective equipment, including lab coats, gloves, and safety goggles.

Handle acids with care and follow safety protocols for their use.

3.8 Data Analysis:

Analyze the ICP-MS data to determine the concentration of mercury in each bone sample.

4.0 Reporting:

Prepare a detailed report including sample information, digestion process, ICP-MS analysis results, and any relevant observations.

5.0 Storage:

Store the digested samples in a designated hazardous waste container for disposal.
