SOP FOR COLONY COUNTER | STANDARD OPERATING PROCEDURE FOR CLEANING, OPERATION & CALIBRATION OF COLONY COUNTER

  

1.0              OBJECTIVE

To lay down the procedure for cleaning, operation & calibration of the colony counter.

2.0              SCOPE

This SOP shall be applicable for cleaning, operation & calibration of the colony counter in the Microbiology Laboratory.

3.0              RESPONSIBILITY





3.1              Microbiologists and above shall be responsible for the preparation and execution of this SOP

3.2              QC & QA Head shall be responsible for the checking and approval of this SOP.

5.0              PROCEDURE

5.1              Cleaning

5.1.1        Ensure that the power supply to the colony counter is switched OFF.

5.1.2        Colony counter shall be dedust daily externally with a clean dry cloth or 70% I.P.A.

5.2              Operation

5.2.1        Ensure that the colony counter r is properly connected to the power supply.

5.2.2        Now Switch  'ON'  the main switch of the equipment.

5.2.3        The digital display shall show “0000” if the digital display is not showing “0000” then press the reset button, digital the display will show “0000”.

5.2.4        A petri dish shall be placed on the illuminating surface.

5.2.5        Ensure that the magnifying glass is adjusted to view a clear colony present on the Petri dish.

5.2.6        The lid of the marker pen shall be open and properly press the tip of the pen on the colonies present on the petri dish and ensure that the digital display shall show you a colony in numbers.

5.3              Calibration of the Instrument:

5.3.1        Ensure for the operating procedure and after that calibration shall be started.

5.3.2        An empty petri dish shall be placed on the illuminating surface for calibration.

5.3.3        The tip of marker pen shall be press 5 times on petri dish and then ensure the number displayed on the screen, it shall be 0005.

5.3.4        The RESET button shall be press and the digital display will show “0000”.

5.3.5        The same procedure shall be repeated by pressing the marker pen 10 times, 15 times, 20 times, 25 times, 50 times, and 100 times.

5.3.6        Ensure that the calibration of colony counter is valid if the number displayed on the screen is equal to the number of times of marker pen pressed.

5.3.7        After completion of work the reset button shall be press and switch off the mains.

5.3.8        Frequency: once in a month
