STANDARD OPERATING PROCEDURE FOR OPERATION OF COMPOUND MICROSCOPE

1.0              OBJECTIVE

To lay down the procedure for the operation of a compound microscope.

2.0              SCOPE

This SOP shall be applicable for the operation of the compound microscope in the Microbiology Lab.

3.0              RESPONSIBILITY

3.1              Microbiologists and above shall be responsible for the preparation and execution of this SOP

3.2              QC & QA Head shall be responsible for the checking and approval of this SOP.

5.0              PROCEDURE

5.1              Observation and viewing of objects

5.1.1           Microscope shall be kept on a vibration-free platform.

5.1.2           Surface of microscope shall be clean with a clean cotton cloth to remove the dust.

5.1.3           Eyepiece, magnifying lenses and condenser shall be clean with cotton soaked in xylene.

5.1.4           Frequency: Before and after use.

5.1.5           Ensure that the power supply to the microscope is switched ON so that the bulb will glow.

5.1.6           Low power (10x) objective shall be adjusted by rotating the revolving nose piece in position.

5.1.7          The focus of light shall be adjusting with the help of a mirror and condenser.

5.1.8         When the image comes into focus with the low power (10x) objective, the nose piece is rotated to the next lens (i.e, 40x or 100x) by simply swinging the objective desire into place, without raising the body tube.

5.1.9          When using the oil immersion objective, the high power objective shall be swing partially out of way and place a drop of immersion oil on the area of the slide where observing and bring the oil- immersion objective into position.

5.1.10       Now look through the eyepiece and focus the object with a fine-adjustment knob.

5.1.11       A slide can be moved in left, right, upper and lower direction to observe the complete smear area with the help of the slide stage screw and coarse adjustment.

5.1.12       After completion of observation slides shall be removed, objective lens shall be clean and condenser with cotton soaked in xylene. Lower down the condenser and place low power objective in position.

5.2                Precautions

5.2.1           Slide shall be observing with both eyes open to have less strain on your eyes.

5.2.2           Always focus your slide slowly and carefully and with low- power objective first

5.2.3           Ensure for keeping the microscope and its parts (e.g stage, lenses, mirror) clean and handle all the parts with care.

5.2.4           Microscope shall be covered always when not in use to keep out dust.

5.2.5           On the completion of the exercise, the microscope shall be cleaned thoroughly.
