1.0 OBJECTIVE
To lay down the procedure for operation of Refractometer
2.0
SCOPE
This SOP shall be applicable for cleaning, operation
& calibration of Refractometer in Chemistry Lab.

RESPONSIBILITY
3.1
Chemist and above
shall be responsible for the preparation and execution of this SOP
3.2
QC & QA Head shall be responsible for the checking and approval of this
SOP.

5.0
PROCEDURE
5.1
Cleaning
5.1.1
Ensure that the eye lens, mirror, and prisms shall be clean.
5.1.2
Ensure that the instrument shall be free from dust.
5.1.3
Platform of Refractometer shall be wiped with a dry lint the free cloth on a daily basis so that there will be no dust particles on the Refractometer.
5.2
Operation
5.2.1
Ensure that the instrument shall be kept in a place where
adequate light reflects
5.2.2
Ensure that the mirror shall be a trough.
5.2.3
Ensure that the prism lock shall be open and smear a drop of
distilled water at 20 oC (use Thermometer and water jacket with
tubes provided in the instrument).
5.2.4
Ensure that the prism lock shall be closed.
5.2.5
Ensure that the eyepiece cross wire shall be adjusted using
knobby rotating it clockwise or anticlockwise (Simultaneously view through
right eyepiece) so that a sharp demarcation line passes through its center (it
is produced in the form of a circle having half light and half dark portion).
5.2.6
Ensure that the fine adjustment shall be done by a knob
provided just above the prism.
5.2.7
Ensure that observe the scale reading (view through left
eyepiece) which gives the refractive index of water at 20 o C (1.333).
5.2.8
Ensure that the prism lock shall be open and dry the prism
with tissue paper.
5.2.9
Ensure that at specified temperature smear a drop of sample the solution shall be on the prism and close the prism lock.
5.2.10 Ensure that the eyepiece
crosswire shall be adjust using the knob by rotating it clockwise or
anticlockwise, so that the Sharp demarcation line passes through its center.
5.2.11 Ensure that the scale
reading shall be observed which gives the refractive index of the sample solution.
5.2.12 Ensure that the instrument
shall be clean after use and switch “OFF” the lamp.
