To lay down a procedure to be followed for sterilization
of experimental tools.

SCOPE
2.1
This Standard Operating Procedure (SOP) shall be applicable for sterilization of
experimental tools in Animal House Facility.

PROCEDURE
5.1
Equipment for animals in the experiment should be free from microbial contamination. Before the use of any
equipment for the animal experiment it should be properly sterilized in
autoclave.
5.2
Material Preparation
5.2.1
Ensure
that the material is safe for autoclaving.
5.2.2
Samples
containing solvents or substances that may emit toxic fumes shall not be
autoclaved.
5.2.3
Glassware
shall be inspected for cracks prior to autoclaving.
5.2.4
Material
shall be prepared and packaged suitably:

Loose
dry materials shall be wrapped or bagged in steam-penetrating paper or loosely covered
with aluminum foil. Wrapping too tightly may impede steam penetration,
decreasing the effectiveness of the process.
5.2.6
All lids
shall be loosened to prevent pressure buildup. All containers shall be covered
by a loosened lid or steam-penetrating bung.
5.2.7
Heat-resistant
borosilicate glassware/s shall be used.
5.2.8
Plastics
must be heat-resistant, i.e., polycarbonate (PC), PTFE (“Teflon”) and most
polypropylene (PP) items.
5.2.9
All
items shall be tagged with autoclave tape.
5.2.10
Items
shall be placed in a stainless steel pan or other autoclavable container for their stability and ease of handling.
5.2.11
Bags
must not be tightly sealed as steam cannot penetrate.
5.3
Autoclave loading and operation
5.3.1
Autoclave
loading and use shall be done as per respective microbiology laboratory SOP.
5.3.2
Note: Do not mix incompatible materials.
Don’t overload and leave sufficient room for
steam circulation. If necessary, keep the container on its side to maximize
steam penetration and avoid entrapment of air.
5.4
After
completion of the sterilization cycle keeps the equipment at a clean dry and preferably sterile place with intact wrapping.
5.5
Remove
the wrapping immediately before use.
