STANDARD OPERATING PROCEDURE FOR VALIDATION OF UV LIGHT STERILIZATION


1.0 OBJECTIVE

To lay down the procedure for validation of U.V light sterilization.

2.0 SCOPE

This SOP shall be applicable for validation of U.V light sterilization in the Microbiology Lab.

3.0 RESPONSIBILITY

3.1 Microbiologist and above shall be responsible for the preparation and execution of this SOP

3.2 QC & QA Head shall be responsible for the checking and approval of this SOP.

3.3 HOD shall be responsible for the authorization of this SOP.

5.0 PROCEDURE

5.1 Soyabean Casein Digest Agar plates shall be prepared as per SOP No………………..

5.2 Ensure that four soybean casein digest agar plates shall be taken and inoculated with 10-20 cfu of Bacillus subtilis, E.coli, S.aureus, etc.

5.3 Out of these four plates, two plates shall be incubating at 35°- 37°C for 48 hours and marked as control, and the remaining two plates shall be exposed under UV light which is to be validated.

5.4 Exposure time shall be given for one hour by keeping UV Light ON.

5.5 After that the plates shall be incubated at 35°- 37° C for 72 hours.

5.6 Observations of plates shall be done.

5.7 Acceptance Criteria

The number of colonies observed in plates exposed to U V should be 10 times less than the number of colonies observed in control/ unexposed plates.

Frequency: After every 100 burning hours of U V Light.
