1.0 PURPOSE
To design the standard operating procedure for the operation of Gel Electrophoresis.
2.0 SCOPE
2.1 This SOP shall be applicable for the operation of Gel Electrophoresis.
3.0 RESPONSIBILITY
3.1 All the users including Scientists and Research associates of the Molecular biology department are responsible for the implementation of this SOP and maintaining the Gel Electrophoresis.
4.0 DISTRIBUTION
4.1 The Quality Assurance department is responsible to keep SOP ‘Master Copy’ approved through the Quality Manager.
4.2 The copy of ‘Control Copy’ of this SOP is being distributed in the biology department and placed near related Equipment/Instruments as ‘Display Copy’.

6.1 Definitions
6.1.1 Gel electrophoresis is a method for the separation and analysis of macromolecules (DNA, RNA, and proteins) and their fragments, based on their size and charge. It is used in clinical chemistry to separate proteins by charge and/or size (IEF agarose, essentially size-independent) and in biochemistry and molecular biology to separate a mixed population of DNA and RNA fragments by length, to estimate the size of DNA and RNA fragments or to separate proteins by charge.
7.0 PROCEDURE
7.1 Fill in the gel running buffer in the electrophoresis tank.
7.2 Cast the agarose gel and set the electrophoresis assembly. Ensure to connect the electrodes in the proper places.
7.3 Turn on the power supply and set the desired current, voltage, and time.
7.4 Turn off the power supply after use.
8.0 PRECAUTIONS
8.1 Always wear protective gloves, safety glasses & a lab coat while preparing & casting agarose gels.
8.2 Ethidium bromide is a suspected carcinogen & should be handled with extreme care. Dispose of used EtBr solutions & gels appropriately.
8.3 Do not use abrasive or highly alkaline cleaners on electrophoresis assembly.
