The purpose of the document is to establish a general and uniform method for the determination of the fructose-glucose ratio in honey.

This standard testing procedure is suitable for the determination of fructose- glucose ratio in honey.

4.0 REQUIREMENTS

4.2 Glassware
4.1.1 Iodine Flask-250 ml
4.1.2 Burette-50 ml
4.1.3 Pipett-25 ml,50 ml
4.1.4 Volumetric Flask-250 ml
4.3 Reagent
4.2.1 Iodine solution: 0.05 N
4.2.2 Sodium Hydroxide solution: 0.1 N
4.2.3 Conc. Sulphuric Acid
4.2.4 Sodium thiosulphate solution: 0.05 N
5.0 PROCEDURE
5.1 Take 50 ml of the prepared solution for total reducing sugar in a 250 ml Iodine flask with the help of a pipette.
5.2 Add 40 ml Iodine solution(0.05 N) in flask.
5.3 Add 25 ml 0.1N NaOH solution in a flask and place in the dark for 20 min.
5.4 Acidify with 5 ml H2SO4 (conc.)
5.5 Titrate the excess of iodine against standard (0.05N) sodium thiosulphate solution.
5.6 Color disappears and notes down the reading.
5.7 Conduct a blank with 50 ml water in place of the sample solution.
