1.0 OBJECTIVE: To design Standard Operating Procedure for Fumigation.
2.0 RESPONSIBILITY: This is the responsibility of the Animal House In-charge, Facility Supervisor and Manager, and Animal Care Persons.
3.0 PRINCIPLE:
Formaldehyde fumigation is a simple and easy procedure to perform but is inherently hazardous if practiced without appropriate precautions. Formaldehyde vapor is an extremely effective biocidal agent. It acts as an alkylating agent, inactivating micro-organisms by reacting with carboxyl, amino, hydroxyl, and sulfhydryl groups of proteins as well as amino groups of nucleic acid bases.
4.0 REQUIREMENT:
4.1 Vaporize about 35ml of Formalin (40% formaldehyde solution) for each cubic meter of enclosed space.
4.2 The inlet, outlet, and any other open spaces are sealed and covered using adhesive tape.
4.3 Large stainless steel bowl / plastic container/glass bottles.
4.4 Potassium permanganate (KMnO4) solid.
4.5 Masks, gloves, and goggles.
5.0 PROCEDURE:
5.1 Fumigation:
5.1.1 Suitable size of fumigation container is placed in the area.
5.1.2 The formaldehyde solution is placed in the container; potassium permanganate is added to the container.
5.1.3 The door is locked and the “FUMIGATION IN PROGRESS – DO NOT OPEN”, board is put on.
5.1.4 Fumigation is done for approximately 24 hours.
5.2 De-fumigation:
5.2.1 After the stipulated duration (24 hours or overnight), the exhaust fan is opened and it is ensured that no traces of formaldehyde are left.
5.2.2 Dilute Ammonium solution is used to absorb the fumes & the entire area is cleaned thoroughly before the start of the activities with mild detergents & disinfectants.
5.2.3 After fumigation the plates are settled and exposed to ascertain the environmental microbial load of the area.
5.2.4 The microbial load at any given place should not exceed the alert limit of 2 x105.
5.2.5 If the violet color does not persist then de-fumigation is complete.
5.2.6 The log sheets should be maintained for fumigation & de-fumigation in their respective log sheets.
5.3 Frequency: Every three month
6.0 PRECAUTION:
6.1.1 Ensure that all Raw materials, Intermediate Products & Primary Packaging Materials in the fumigation area are closed properly, if applicable.
6.1.2 All the doors, windows, and any other openings should be sealed properly.
NOTE: Formalin is a strong irritant & is toxic. Inhalation of formaldehyde gas should be avoided since it causes irritation of the respiratory tract, and contact with the skin should also be avoided.
6.1.3 Personnel must leave the area being fumigated as soon as possible after fumigation.
6.1.4 Fumigation should be supervised by responsible personnel.
6.1.5 To insist on the operation in the event of an accident.
6.1.6 Persons coming in contact with formaldehyde solution must wash their hands and faces thoroughly with soap and water before eating &/ or drinking.
6.1.7 The air blowers/ slit air conditioners, U.V. lamps, and exhaust fans are switched off.
