PURPOSE:
This standard operating procedure (SOP) outlines the procedures for conducting rodent surgeries in a safe, efficient, and ethical manner. The purpose of this SOP is to ensure that all personnel involved in rodent surgeries follow standardized procedures to minimize the risk of complications, infection, and stress on the animals.
RESPONSIBILITY:
The Principal Investigator (PI) is responsible for ensuring that all personnel involved in rodent surgeries have received appropriate training and follow this SOP. The PI must also ensure that the Institutional Animal Care and Use Committee (IACUC) has approved the surgical procedures and that all surgeries are conducted in compliance with animal welfare regulations.
MATERIALS:
Surgical instruments (e.g., scalpel, forceps, scissors, suture material, wound clips, autoclips)
Sterile surgical drapes, gauze, gowns, gloves, and masks
Anesthesia machine and supplies (e.g., isoflurane, oxygen)
Sterile saline isotonic solution for injection (e.g. 0.9%saline) and sterile ophthalmic ointment or other sterile fluids for hydration and rinsing (e.g. 70% Alcohol)
A heating disc, warming pad, or warm-water circulating pad (do not use electric heating pads)
Suture materials and wound closure supplies (e.g. Chlorhexidine 2% solution or povidone-iodine solution)
Analgesics and other supportive care supplies
Electric Trimmer
Dry bead sterilizer or cold sterilization agents (e.g. glutaraldehyde) and 70% alcohol (as a rinsing agent)
PROCEDURES:
Pre-operative Care:
All rodents should be fasted for a specified period before surgery to prevent vomiting and aspiration during anesthesia.
Rodents should be provided with appropriate bedding, food, and water to maintain their health and well-being.
The surgical site should be shaved and cleaned with a surgical scrub solution.
Animals should be anesthetized using appropriate anesthetic agents and dosages, and anesthesia should be maintained throughout the surgical procedure.
Surgical Principles/Aseptic technique:
All surgical instruments and supplies must be sterile.
The surgical site should be draped with sterile surgical drapes to maintain aseptic conditions.
Personnel should wear sterile surgical gowns, gloves, and masks to minimize the risk of infection.
The surgical procedure should be conducted using an aseptic technique to minimize the risk of contamination.
Surgical Monitoring and Supportive Care:
Vital signs (e.g., heart rate, respiratory rate, body temperature) should be monitored throughout the surgical procedure.
Fluid therapy should be provided to maintain hydration and electrolyte balance.
Analgesics and other supportive care should be provided as needed to manage pain and minimize stress.
Post-operative Care:
Animals should be closely monitored during recovery from anesthesia.
Animals should be kept warm and provided with appropriate post-operative care, including analgesics, fluid therapy, and wound care.
Animals should be observed for signs of pain, distress, or complications, and appropriate interventions provided as needed.
Animals should be housed in appropriate post-operative conditions to promote healing and minimize the risk of complications.
In conclusion, this SOP outlines the procedures for conducting rodent surgeries in a safe, efficient, and ethical manner. It is the responsibility of all personnel involved in rodent surgeries to follow these procedures to minimize the risk of complications and ensure the welfare of the animals.
