SOP FOR PROTECTING PEOPLE AND ANIMALS: WORKING WITH BSL-2 BIOHAZARDS IN RODENT STUDIES

1.0 Purpose:

This SOP outlines how to house and care for rodents exposed to biosafety level 2 (BSL2) biohazards. It helps to:

  • Contain the biohazards
  • Reduce the risk of exposure for people and other animals

2.0 Responsibilities:

  • Principal investigator:
    • Provides information about the biohazard in the Animal Study Protocol (ASP)
    • Gives a copy of the biosafety certificate to the Facility Supervisor
    • Makes sure research staff is trained before doing BSL2 experiments
  • Test Facility Manager:
    • Make sure the animal room and equipment are suitable for BSL2 experiments
    • Provides necessary materials
    • Makes sure staff and users are trained properly
    • Posts information for identifying risks
  • Research and animal care staff:
    • Follows procedures to minimize risks as described in this SOP

3.0 Materials:

  • Personal protective equipment (PPE): gown, gloves, sleeve covers, shoe covers, bonnet, mask, goggles or safety glasses
  • Biological Safety Cabinet (BSC)
  • Microisolator cages
  • Biohazard MSDS
  • Biohazard door sign
  • Biohazard waste containers/bags
  • Sharps container
  • Disinfectant
  • Dedicated materials and equipment (notebooks, cage cards, pens, ear punchers, instruments)

4.0 Procedures:

  • Wear proper clothing: long pants, socks, closed shoes.
  • Wear PPE when handling infectious materials or animals:
    • Bonnet covering all hair
    • Mask covering mouth and nose
    • Disposable gown, tied in the back
    • Double gloves, stretched over gown cuffs
    • Disposable sleeve covers, over gloves
    • Protective eyewear (recommended)
  • Use dedicated materials and equipment.
  • House animals in micro isolator cages or isolators.
  • Label cages with a biohazard sign, biohazard name, and administration date.
  • Do all procedures in a BSC (changing cages, infecting animals, necropsy, etc.).
  • Decontaminate work surfaces after use or spills with disinfectant.
  • Don't eat, drink, or store food in animal rooms.
  • Wash or sanitize hands after handling infectious materials or animals, before leaving the room.
  • Minimize creating aerosols during procedures.
  • Don't recap needles; dispose of them in a sharps container.
  • Have a vermin control program in BSL2 animal rooms and the rest of the facility.

5.0 Changing cages:

  • Do it in the BSC. Dispose of feed, water, and bedding as biohazard waste.
  • Bag dirty cages in an autoclavable bag under the BSC.
  • Seal the bag with autoclavable bands or ties before removing it.
  • Spray the bag with disinfectant before moving it to a second autoclavable bag.
  • Seal the second bag with bands or ties and spray with disinfectant.
  • Autoclave the cages and check for proper steam penetration.
  • Wait for weekly biological indicator results.
  • After autoclaving, dispose of bedding as regular waste and wash cages according to SOP.

6.0 Animal room:

  • Keep doors closed when infected animals are present.
  • Set the room to negative pressure compared to the corridor or anteroom.
  • Limit access to authorized personnel and users.
  • The facility manager must:
    • Post a biohazard sign with the biohazard name, medical requirements (if any), contact information, and PPE requirements.
    • Make sure personnel and users are properly informed about the biohazard and procedures.
  • Dispose of infectious waste and animal carcasses in double biohazard bags or containers.
