SOP FOR GC-MS/MS

 1.   Objective: To lay down the procedure for the operation of GC-MS/MS.

2. Responsibility: It is the responsibility of the Technical Assistant, Technical Manager, Quality Manager, and all those individuals who are working on the instrument.

3.   Instrument Details:

4.  Purpose: This SOP provides an authorized procedure for the operation and maintenance of GC-MS/MS.

5.   Scope: This SOP covers the procedure for the operation and maintenance of GC-MS/MS. 

6.  Principle: GCMSMS is a technique by which a mixture of samples is separated into components using a capillary column meant for gas chromatography coated with a suitable phase and eluting the separated components by employing a suitable gas mobile phase and variation of temperature.

     Before you turn on the MS:

·     Carrier gas must be at least 99.999% purity is plumbed to GC with the recommended.

·     A capillary column is installed in the GC inlet and in the GC-MS interface.

7.   Procedure: 

·     Enter in the log book.

·     Switch on the gases (Helium and Nitrogen) from the cylinder and wall panel. Make sure that cylinder outlet pressure is between 4 - 6 kg.

·     Make sure the instrument is on an un-uninterrupted power supply.

·     Power on the instrument through GC and MS power buttons.

·     Switch on the PC and launch software from the icon

·     Wait for 5 minutes and then the load conditioning method.

·     Keep the system on conditioning for 4 hrs.

·   Load the method in which you want to analyze the method. Set the ALS, Inlets, column, oven, Aux heater, configuration, and readiness as desired. The method file can be saved by assigning a suitable name based on the type of analysis to be undertaken in the method file for easy retrieval at a later time or date. This would help in saving time setting the parameters of the system required for performing the analysis by the click of the method download button. Allow the system to attain the required parameters for the analysis to be performed.

·    In the instrument control panel, click the MS tune icon to display the GC-QQQ Tune dialogue box.

·   Make sure "EI hisenseautotune" is selected. Click on Autotune tab. Tuning will start automatically. Click ok

·    Do Check Tune by clicking on the check tune tab.

·  Make sure each parameter is ok. Click close. Again click on "close" and then "ok" to go to instrument control.

·     For injecting a single sample, put the sample on desired vial position of the autosampler. Also,

         make sure the rinsing solvent is filled in wash vials. Click on the arrow of the run method.

·     Enter data path, data file name, sample name, and vial no. Click on "ok and run method".

·     For running sequence, Click on sequence and Edit sequence:

·     Make desired lines, enter sample name, vial no, method path, Method, Data path, Data file name, and sample type. Click ok.

·     Make sure the data file name is different in each line otherwise, data will be overwritten.

·     Click run sequence under the sequence tab.

·     Enter the data file directory and click "run".
